Indications
- Relief of symptoms associated with seasonal allergic rhinitis
- Relief of symptoms associated with perennial allergic rhinitis
- Treatment of uncomplicated skin manifestations of chronic idiopathic urticaria
- Treatment of allergen induced asthma
Pharmacology
- Potent H1 receptor antagonist
- No significant anticholinergic effects
- No significant antiserotonic effects
- Inhibits histamine-mediated early phase of allergic reaction
- Reduces migration of inflammatory cells
- Reduces release of mediators associated with late phase of allergic reaction
- Peak plasma concentrations of 257 mcg/L within one hour of administration
- Peak plasma concentrations of 980 mcg/L in children
- Almost no drowsiness effect
- Does not cause behavioral changes
- Plasma half-life of approximately 11 hours
- Renal clearance of 30 ml/minute
- Excretion half-life of approximately nine hours
- Consistent absorption from one subject to the next
- No effect of food on extent of absorption
Dosage & Administration
- Adult dosage: 1 tablet or 2 teaspoonfuls daily
- Children 6 years and older dosage: 1 tablet or 2 teaspoonfuls daily
- Children 2-6 years dosage: 1 teaspoonful once daily or 1/2 teaspoonful twice daily
- Children 6 months-2 years dosage: 1/2 teaspoonful once daily
- Cetirizine injectable dosage form for intravenous administration only
- Adult intravenous dosage: 10 mg
- Adolescent intravenous dosage: 5 mg or 10 mg depending on symptom severity
- Child intravenous dosage: 2.5 mg
- Administered once every 24 hours as needed for treatment of acute urticaria

- No clinically significant drug interactions with Theophylline
- No clinically significant drug interactions with Azithromycin
- No clinically significant drug interactions with Pseudoephedrine
- No clinically significant drug interactions with Ketoconazole
- No clinically significant drug interactions with Erythromycin
- No clinically significant drug interactions with other drugs
Contraindications
- Contraindicated in patients with history of hypersensitivity to Cetirizine
- Contraindicated in patients with history of hypersensitivity to hydroxyzine
Side Effects
- Common side effect: somnolence
Pregnancy & Lactation
- US FDA Pregnancy Category B
- No adequate and well-controlled studies in pregnant women
- Use during pregnancy only if clearly needed
- Excretion in human milk
- Caution in administration to a nursing woman
Precautions & Warnings
- Caution when driving a car
- Caution when operating heavy machinery
Use in Special Populations
- Reduced clearance in patients with mild to moderate hepatic impairment
- Reduced clearance in patients with mild to moderate renal impairment
- AUC and half-life increased by about 2 to 3 fold in patients with hepatic or renal insufficiency
- Plasma elimination half-life of approximately 20 hours in dialysis patients
- Safety demonstrated in pediatric patients aged 6 months to 5 years
- Reduced clearance in elderly patients
