Indications
- Adjunct to diet to reduce elevated total cholesterol
- Reduce elevated LDL cholesterol levels
- Reduce apolipoprotein B (Apo-B) levels
- Reduce triglycerides levels
Pharmacology
- Selective inhibitor of HMG-CoA reductase
- Lowers plasma cholesterol and lipoprotein levels
- Inhibits cholesterol synthesis in the liver
- Increases hepatic LDL receptors for enhanced uptake and catabolism of LDL
Dosage & Administration
- Adults: Usually 10 mg once daily; max. 80 mg once daily
- Child (10-18 years): Initially 10 mg once daily, increased if necessary at intervals of at least 4 weeks to max. 20 mg once daily

Contraindications
- Hypersensitivity to any component
- Active liver disease
- Unexplained persistent elevations of serum transaminases
- History of serious adverse reaction to prior HMG-CoA reductase inhibitors

Pregnancy & Lactation
- Contraindicated during pregnancy

Precautions & Warnings
- Liver function tests should be performed before initiation of treatment and periodically thereafter
- Use with caution in patients consuming substantial quantities of alcohol or with a history of liver disease
- Discontinue if markedly elevated CPK levels occur or myopathy is diagnosed or suspected
Use in Special Populations
- Higher plasma concentrations in healthy elderly subjects
- No clinically significant difference in LDL-C reduction between men and women
- Renal disease has no influence on plasma concentrations or LDL-C reduction
- In patients with chronic alcoholic liver disease, plasma concentrations are markedly increased
Overdose Effects
- Specific treatment not available
- Symptomatic and supportive measures should be instituted
- Liver function tests and serum CK levels should be monitored
