Description

  • Indications:
    • Reduction of total cholesterol and LDL cholesterol in patients with familial hypercholesterolaemia
    • Reduction of cholesterol and triglycerides in patients with mixed dyslipidemia
    • Treatment of patients with elevated triglyceride levels in hypertriglyceridaemia
    • Treatment of patients with dysbetalipoproteinaemia
    • Reduction of cardiac ischaemic events in patients with coronary artery disease
    • Reducing hypercholesterolemia associated with diabetes mellitus or renal transplantation
  • Pharmacology:
    • Selective inhibitor of HMG-CoA reductase
    • Lowers plasma cholesterol and lipoprotein levels
    • Increases hepatic LDL receptors for enhanced uptake of LDL
  • Dosage & Administration:
    • Adults: 10-80 mg once daily
    • Children (10-18 years): 10-20 mg once daily
    • Initial dose of 10 or 20 mg once daily for adults
    • Dosage range of 10 to 80 mg once daily
  • Interaction:
    • Increased risk of myopathy with concurrent administration of certain drugs
    • Altered plasma concentrations and response with coadministration of different drugs
  • Contraindications:
    • Hypersensitivity to any component of the medication
    • Active liver disease or unexplained persistent elevations of serum transaminases
    • History of serious adverse reaction to prior administration of HMG-CoA reductase inhibitors

    • Back pain
    • Rash
    • Asthenia
    • Arthralgia

  • Pregnancy & Lactation:
    • Contraindicated during pregnancy
    • Not known if excreted in human milk
    • Maternal treatment may reduce fetal levels of mevalonate
    • Not recommended during breastfeeding
  • Precautions & Warnings:
    • Liver function tests should be performed before initiation of treatment
    • Discontinue therapy if markedly elevated CPK levels occur or myopathy is diagnosed or suspected
  • Use in Special Populations:
    • Higher plasma concentrations in elderly subjects
    • No clinically significant difference in LDL-C reduction between men and women
    • Renal disease has no influence on plasma concentrations or LDL-C reduction
  • Overdose Effects:
    • Specific treatment not available
    • Symptomatic and supportive measures should be instituted
    • Liver function tests and serum CK levels should be monitored
