Description
Lipitin is indicated as an adjunct to diet to reduce elevated total cholesterol, LDL cholesterol, apolipoprotein B (Apo-B) and triglycerides levels in various diseases when response to diet and other non-pharmacological measures is inadequate.
Dosage
- 10 mg once daily for adults
- Increased at intervals of at least 4 weeks to max. 80 mg once daily if necessary
Pharmacology
- Mode of action: Selective inhibitor of HMG-CoA reductase
- Absorption: Rapidly absorbed after oral administration
- Distribution: Mean volume of distribution is approximately 381 liters
- Metabolism: Extensively metabolized to ortho- and parahydroxylated derivatives
- Excretion: Eliminated primarily in bile following hepatic and/or extra-hepatic metabolism
Interaction
- Concurrent administration of cyclosporine, fibric acid derivatives, niacin, erythromycin, azole antifungals increases risk of myopathy
- Coadministration with antacid leads to decreased plasma concentrations
- Coadministration with oral contraceptives increases AUC values for norethindrone and ethinyl estradiol
Contraindications
Hypersensitivity to any component of the medication, active liver disease, unexplained persistent elevations of serum transaminases

Pregnancy lactation
- Pregnancy: Contraindicated. No controlled clinical trials in pregnant women.

Precautions warnings
Liver function tests should be performed before the initiation of treatment and periodically thereafter.
Special populations
- Geriatric: Plasma concentrations are higher in healthy elderly subjects
- Pediatric: Pharmacokinetic data not available
- Renal insufficiency: No influence on the plasma concentrations of the drug
- Hepatic insufficiency: Plasma concentrations are markedly increased in patients with chronic alcoholic liver disease
Overdose effects
Specific treatment not available. Patient should be treated symptomatically and supportive measures instituted.
