Usage and Dosage
- Lipitin is used to reduce total cholesterol and LDL cholesterol in patients with familial hypercholesterolaemia
- For the treatment of patients with elevated serum triglyceride levels in hypertriglyceridaemia
- To reduce cardiac ischaemic events in patients with coronary artery disease with elevated LDL-cholesterol level
Pharmacology
- Atorvastatin is a selective inhibitor of HMG-CoA reductase responsible for cholesterol synthesis
Absorption
- Rapid absorption after oral administration with maximum plasma concentrations occurring within 1 to 2 hours
Distribution
- Mean volume of distribution is approximately 381 liters
- Atorvastatin is 98% bound to plasma proteins
Metabolism
- Extensively metabolized to ortho- and parahydroxylated derivatives and various beta-oxidation products
- Metabolized by cytochrome P450 3A4
Excretion
- Eliminated primarily in bile following hepatic and/or extra-hepatic metabolism
Side Effects
- Frequent side effects include constipation, flatulence, and dyspepsia
- Other side effects may include infection, headache, back pain, and rash
Pregnancy & Lactation
- Atorvastatin is contraindicated during pregnancy
- It is not known whether atorvastatin or its metabolites are excreted in human milk
Precautions & Warnings
- Liver function tests should be performed before initiation of treatment and periodically thereafter
Use in Special Populations
- Plasma concentrations are higher in healthy elderly subjects than in young adults
- Pharmacokinetic data in the pediatric population are not available
Overdose Effects
- Specific treatment is not available for atorvastatin overdose
