Indications
- Relief from seasonal allergic rhinitis
- Relief from perennial allergic rhinitis
- Relief from skin allergies including chronic urticaria
- Alleviates symptoms of allergic rhinitis like sneezing, nasal discharge, itching, ocular itching and burning
- Indicated in idiopathic urticaria
- Symptomatic relief of seasonal allergic rhinitis in children over 2 years
- Relief from allergic skin conditions in children over 2 years
Pharmacology
- Preparation of Loratadine
- Non-sedative histamine H1 receptor antagonist with antiallergic properties
- Long-acting tricyclic antihistamine
- Selective peripheral Hi-receptor antagonistic activity
- Rapidly effective and long-lasting
- Allows once-a-day administration
Dosage and Administration
- Adults and children over 12 years: One tablet once daily
- Children 2-12 years: Body weight over 30 kg - one tablet once daily, below 30 kg - half tablet once daily
- Not recommended for use in children below 2 years of age
Interaction
- No reports of potentially hazardous interactions with other drugs
- No potentiating effects when administered concurrently with alcohol
- Concomitant therapy with drugs inhibiting or metabolized by hepatic cytochromes P450 3A4 and 2D6 may elevate plasma concentrations
- Cimetidine inhibits both enzymes, erythromycin or ketoconazole inhibits P450 3A4
- Increased serum concentrations reported with no adverse effects
Contraindications
- Hypersensitivity or idiosyncrasy to components
Side Effects
- Comparable incidence of adverse events to placebo during clinical studies
- No significant sedation and anticholinergic effects observed with therapeutic dose
- Comparable effect on actual driving performance, cognitive and psychomotor functioning to placebo
Pregnancy and Lactation
- Not advisable during pregnancy due to lack of experience of use in human pregnancy
- Excreted in breast milk in very small amount
- Nursing mothers advised not to take the drug
Precautions and Warnings
- Caution in patients with liver impairment or renal insufficiency (eGFR <30 ml/min)
Overdose Effects
- Symptoms in adults: somnolence, tachycardia, headache with overdose greater than 10 mg
- Symptoms in children: extrapyramidal signs, palpitations with overdose greater than 10 mg
- Prompt symptomatic and supportive measures should be instituted in case of overdosage
- Early treatment with oral activated charcoal in large overdoses
- Inducing vomiting or gastric lavage may be performed in conscious patients
