Dosage & Administration
- Adult dosage: Once daily
- Administer in the morning
- Children 2-12 years dosage based on body weight
- Not recommended for children below 2 years
Interaction
- No hazardous interactions with other drugs
- No potentiating effects when administered with alcohol
- Potential elevation of plasma concentrations with hepatic cytochromes P450 3A4 and 2D6 inhibitors
- Cimetidine inhibits both enzymes
- Erythromycin or ketoconazole inhibits P450 3A4
- Increase in Loratadine serum concentrations
Contraindications
- Hypersensitivity
- Idiosyncrasy to components
Side Effects
- Comparable adverse events incidence to placebo
- No sedation observed in clinical studies
- Comparable to placebo in driving performance and cognitive functioning
Pregnancy & Lactation
- Not advisable during pregnancy
- Excreted in breast milk in a very small amount
Precautions & Warnings
- Caution in patients with liver impairment
- Caution in patients with renal insufficiency (eGFR <30 ml/min)
Overdose Effects
- Reported effects in adults: somnolence, tachycardia, headache
- Reported effects in children: extrapyramidal signs, palpitations
- Prompt symptomatic and supportive measures
- Consider oral activated charcoal for early presentation after overdose
