Indications
- Relief from symptoms of seasonal allergic rhinitis
- Relief from symptoms of perennial allergic rhinitis
- Relief from symptoms of skin allergies including chronic urticaria
- Alleviating symptoms of allergic rhinitis such as sneezing
- Alleviating symptoms of allergic rhinitis such as nasal discharge
- Alleviating symptoms of allergic rhinitis such as itching
- Alleviating symptoms of allergic rhinitis such as ocular itching
- Indicated in idiopathic urticaria
- Indicated for symptomatic relief of seasonal allergic rhinitis in children over 2 years
- Indicated for symptomatic relief of allergic skin conditions in children over 2 years
Pharmacology
- Preparation contains Loratadine
- Non-sedative histamine H1 receptor antagonist
- Antiallergic properties
- Long-acting tricyclic antihistamine
- Selective peripheral H1-receptor antagonistic activity
- Rapidly effective and long-lasting
- Allows once-a-day administration
Dosage & Administration
- One Loratadine tablet once daily for adults and children over 12 years of age
- Usually administered in the morning or when symptoms require treatment
- Half Loratadine tablet once daily for children 2-12 years and body weight below 30 kg
- Not recommended for use in children below 2 years of age
Interaction
- No reports of potentially hazardous interactions with other drugs
- No potentiating effects when administered concurrently with alcohol
- Concomitant therapy with drugs that inhibit or are metabolized by hepatic cytochromes P450 3A4 and 2D6 may elevate plasma concentrations
- Drugs that inhibit P450 3A4 increase Loratadine serum concentrations
Contraindications
- Hypersensitivity or idiosyncrasy to Loratadine components
Side Effects
- Comparable incidence of adverse events, sedation, and anticholinergic effects with 10 mg Pretin to that observed with placebo
Pregnancy & Lactation
- No experience of the use of Loratadine in human pregnancy
- Not advisable during pregnancy
- Excreted in breast milk in a very small amount
- Nursing mothers advised not to take the drug
Precautions & Warnings
- Caution in patients with liver impairment
- Caution in patients with renal insufficiency (eGFR < 30 ml/min)
Overdose Effects
- Somnolence, tachycardia, and headache reported in adults with overdose > 10 mg
- Extrapyramidal signs and palpitations reported in children with overdose > 10 mg
- Institute general symptomatic and supportive measures promptly and maintain as necessary
- Consider oral activated charcoal for early presentation after large overdoses
- Induce vomiting or perform gastric lavage for conscious patients
