Description
- Indicated for reducing elevated total cholesterol, LDL cholesterol, apolipoprotein B (Apo-B), and triglycerides levels in various diseases
- Selective inhibitor of HMG-CoA reductase
- Rapidly absorbed after oral administration with maximum plasma concentrations within 1-2 hours
- Approximately 98% bound to plasma proteins
- Extensively metabolized to ortho- and parahydroxylated derivatives
- Primarily eliminated in bile following hepatic and/or extra-hepatic metabolism
- Recommended starting dose is 10 or 20 mg once daily, with a dosage range of 10 to 80 mg once daily
- Interacts with drugs like cyclosporine, fibric acid derivatives, niacin, erythromycin, azole antifungals, antacids, colestipol, digoxin, oral contraceptives, and warfarin
- Contraindicated in patients with hypersensitivity to any component, active liver disease, unexplained persistent elevations of serum transaminases, and history of serious adverse reaction
- Common side effects include constipation, flatulence, dyspepsia, abdominal pain, infection, headache, back pain, rash, asthenia, arthralgia, and myalgia
- Not recommended during pregnancy or lactation
- Liver function tests should be performed before the initiation of treatment and periodically thereafter
- Higher plasma concentrations observed in elderly subjects compared to young adults
- No clinically significant difference in LDL-C reduction between men and women
- No dose adjustment necessary in patients with renal dysfunction
- Specific treatment not available for overdose, with supportive measures instituted as required
