Description

  • Taven is indicated to reduce elevated levels of total cholesterol, LDL cholesterol, apolipoprotein B (Apo-B), and triglycerides in various diseases.
  • It is a selective inhibitor of HMG-CoA reductase and lowers plasma cholesterol and lipoprotein levels.

Indications

  • Reduction of total cholesterol and LDL cholesterol in patients with heterozygous and homozygous familial hypercholesterolaemia
  • Reduction of elevated cholesterol and triglycerides in patients with mixed dyslipidemia
  • Treatment of hypertriglyceridaemia and dysbetalipoproteinaemia
  • Reduction of cardiac ischaemic events in patients with coronary artery disease
  • Management of hypercholesterolemia associated with diabetes mellitus or renal transplantation

Pharmacological Action

  • Selective inhibition of HMG-CoA reductase
  • Reduction of cholesterol synthesis in the liver
  • Increase in hepatic LDL receptors for enhanced uptake and catabolism of LDL

Absorption

  • Rapid absorption after oral administration
  • Maximum plasma concentrations occur within 1 to 2 hours

Distribution

  • Mean volume of distribution of approximately 381 liters
  • Approximately 98% bound to plasma proteins

Metabolism

  • Extensive metabolism to active metabolites
  • In vitro studies suggest metabolism by cytochrome P450 3A4

Excretion

  • Primarily eliminated in bile following hepatic and/or extra-hepatic metabolism
  • Mean plasma elimination half-life of approximately 14 hours

Interaction

  • Increased risk of myopathy with concurrent administration of certain drugs
  • Plasma concentration changes when coadministered with antacids or colestipol
  • Effects on plasma concentrations when coadministered with digoxin, erythromycin, oral contraceptives, and warfarin

Contraindications

  • Hypersensitivity to any component of the medication
  • Active liver disease or unexplained persistent elevations of serum transaminases
  • History of serious adverse reaction to prior administration of HMG-CoA reductase inhibitors

  • Higher plasma concentrations in healthy elderly subjects
  • No clinically significant difference in LDL-C reduction between men and women
  • Pharmacokinetic data not available in the pediatric population
