Description
Tigilow 10 mg Tablet is a selective inhibitor of HMG-CoA reductase and is used to reduce elevated total cholesterol, LDL cholesterol, apolipoprotein B (Apo-B), and triglyceride levels in various diseases. It is also used for the prevention of cardiovascular events and to reduce the risk of cardiac ischaemic events. The medicine is generally well-tolerated but may have side effects related to gastrointestinal discomfort and other rare side effects.

Indication
- Reduction of total cholesterol and LDL cholesterol in patients with heterozygous and homozygous familial hypercholesterolaemia
- Treatment of patients with elevated serum triglyceride levels in hypertriglyceridaemia
- Prevention of cardiovascular events in patients with asymptomatic or mild to moderate symptomatic coronary artery disease
Pharmacology
- Mechanism of action: Selective inhibitor of HMG-CoA reductase
- Absorption: Rapidly absorbed after oral administration; maximum plasma concentrations occur within 1 to 2 hours.
- Metabolism: Extensively metabolized to ortho- and parahydroxylated derivatives and various beta-oxidation products.
- Excretion: Eliminated primarily in bile following hepatic and/or extra-hepatic metabolism.
Dosage administration
- Recommended dose: 10 mg once daily
- Max dose: 80 mg once daily
- Dosage adjustment guidelines: Lipid levels should be analyzed within 2 to 4 weeks and dosage adjusted accordingly.
Interactions
- Increased risk of myopathy with concurrent administration of certain drugs
- Plasma concentrations affected when coadministered with antacids, colestipol, digoxin, erythromycin, oral contraceptives, or warfarin

- Hypersensitivity to any component of the medication
- Active liver disease or unexplained persistent elevations of serum transaminases
- History of serious adverse reaction to prior administration of HMG-CoA reductase inhibitors

- Hemodialysis: Not expected to significantly enhance clearance
- Hepatic insufficiency: Plasma concentrations are markedly increased in patients with chronic alcoholic liver disease
