Description
- Vastin 10 mg Tablet is a selective inhibitor of HMG-CoA reductase.
- It is used to avoid heart attacks and Primary Hypercholesterolemia.
- It is also used to treat conditions such as Mixed Dyslipidemia and Hypertriglyceridemia.
Indications
- Reduction of total cholesterol
- Reduction of LDL cholesterol
- Treatment of heterozygous familial hypercholesterolaemia
- Treatment of homozygous familial hypercholesterolaemia
- Reduction of elevated cholesterol and triglycerides in mixed dyslipidemia (Fredrickson Type Ia and Ib)
- Treatment of hypertriglyceridemia (Fredrickson Type IV)
- Treatment of dysbetalipoproteinaemia (Fredrickson Type III)
- Reduction of cardiac ischaemic events in patients with coronary artery disease
- Reduction of cholesterol concentrations in patients with hypercholesterolemia associated with diabetes mellitus or renal transplantation
Pharmacology
- Selective inhibition of HMG-CoA reductase
- Lowering of plasma cholesterol and lipoprotein levels
- Increase in hepatic LDL receptors for enhanced uptake and catabolism of LDL
- Rapid absorption after oral administration
- Extensive metabolism to ortho- and parahydroxylated derivatives
- Mean volume of distribution approximately 381 liters
- Approximately 98% bound to plasma proteins
- Mean plasma elimination half-life approximately 14 hours
- Extent of absorption increases in proportion to dosage
DosageAndAdministration
- Usually 10 mg once daily for adults
- Initially 10 mg once daily for children (10-18 years)
- Increased dose intervals of at least 4 weeks
- Maximum dose of 80 mg once daily for adults
- Usual maximum dose of 20 mg once daily for children (10-18 years)
- Placed on a standard cholesterol-lowering diet before treatment
- Lipid levels analyzed within 2 to 4 weeks of initiation
Interaction
- Increased risk of myopathy with concurrent administration of specific drugs
- Plasma concentration alterations when coadministered with antacids, colestipol, digoxin, erythromycin, oral contraceptives, warfarin
Contraindications
- Hypersensitivity to any component of the medication
- Active liver disease or unexplained persistent elevations of serum transaminases
- History of serious adverse reaction to prior administration of HMG-CoA reductase inhibitors
