Description
- Selective inhibitor of HMG-CoA reductase
- Reduces elevated cholesterol levels

- Recommended starting dose of 10 or 20 mg once daily

- Increases hepatic LDL receptors for enhanced uptake of LDL

- Rapid absorption after oral administration
- Maximum plasma concentrations occur within 1 to 2 hours

- Mean volume of distribution of approximately 381 liters
- 98% bound to plasma proteins
Metabolism
- Extensively metabolized to ortho- and parahydroxylated derivatives and beta-oxidation products
- Importance of metabolism by cytochrome P450 3A4
Excretion
- Eliminated primarily in bile following hepatic and/or extra-hepatic metabolism
- Mean plasma elimination half-life of approximately 14 hours
Interaction
- Concurrent administration with cyclosporine, fibric acid derivatives, or erythromycin increases risk of myopathy
- Altered plasma concentrations with coadministration of antacid
- Increased digoxin concentrations with coadministration of atorvastatin
- Coadministration with oral contraceptives may increase AUC values for certain hormones

- Hypersensitivity to any component of the medication
- Active liver disease
- Unexplained persistent elevations of serum transaminases
- History of serious adverse reaction to prior administration of HMG-CoA reductase inhibitors
