Indications
- Treatment of conjunctivitis
- Treatment of superficial ocular infections
- Adjunctive therapy for trachoma
- Effective against Escherichia coli
- Effective against Staphylococcus aureus
- Effective against Streptococcus pneumoniae
- Effective against Haemophilus influenzae
- Effective against Klebsiella species
- Effective against Enterobacter species
Pharmacology
- Bacteriostatic activity
- Inhibition of bacterial synthesis of dihydrofolic acid
- Competitive inhibition of dihydropteroate synthetase
- Activity against susceptible bacterial eye pathogens
- Inactivity against Neisseria species
- Inactivity against Serratia marcescens
- Inactivity against Pseudomonas aeruginosa
- Resistance in staphylococcal isolates
Dosage & Administration
- Instill one or two drops into the affected eye(s) every two to three hours
- Taper dosages as the condition responds
- Usual treatment duration of seven to ten days for conjunctivitis
- Instill two drops every two hours for trachoma
- Topical administration must be accompanied by systemic administration
Interaction
- Incompatibility with silver preparations
Contraindications
- Hypersensitivity to sulfonamides or any ingredient of the preparation
Side Effects
- Development of bacterial and fungal corneal ulcers
- Local irritation
- Stinging and burning
- Non-specific conjunctivitis
- Conjunctival hyperemia
- Secondary infections
- Allergic reactions
- Rare severe reactions including Stevens-Johnson syndrome
- Rare severe reactions including toxic epidermal necrolysis
- Rare severe reactions including fulminant hepatic necrosis
- Rare severe reactions including agranulocytosis
- Rare severe reactions including aplastic anemia
- Rare severe reactions including other blood dyscrasias
- Rare fatalities
Pregnancy & Lactation
- Pregnancy Category C
- Potential risk to fetus
- Potential development of kernicterus in newborn
- No adequate and well-controlled studies in pregnant women
- Consideration of potential benefit justifying potential risk
- Potential for development of kernicterus in neonates of lactating women
Precautions & Warnings
- Prolonged use may lead to overgrowth of nonsusceptible organisms
- Bacterial resistance may develop
- Reduced effectiveness in presence of para-aminobenzoic acid
- Possible recurrence of sensitization
- Possible cross-sensitivity between different sulfonamides
- Discontinuation in case of hypersensitivity, purulent discharge, or aggravation of inflammation or pain
Use in Special Populations
- Safety and effectiveness not established in infants below two months of age
