Indications
- Treatment of acute clinical manifestations of Herpes simplex virus in immunocompromised patients
- Treatment of severe primary or non-primary genital herpes in immune competent patients
- Treatment of Varicella zoster virus infection in immunocompromised patients
- Treatment of Herpes zoster (shingles) in immune competent patients
- Treatment of Herpes simplex encephalitis
Pharmacology
- Interference with DNA synthesis
- Inhibition of viral replication
- Intracellular conversion to Acyclovir Triphosphate
- Conversion to Acyclovir Monophosphate via virus-coded thymidine kinase
- Phosphorylation to diphosphate via cellular guanylate kinase
Dosage
- Dosage for Herpes simplex infection: 5 mg/kg every 8 hours
- Dosage for severe Herpes zoster infection (shingles): 5 mg/kg every 8 hours
- Dosage for Varicella zoster infection: 10 mg/kg every 8 hours
- Dosage for Herpes simplex encephalitis: 10 mg/kg every 8 hours
- Each dose should be administered by slow intravenous infusion over a one-hour period
Administration
- Recommended duration of administration: 5 to 7 days for most infections
- Recommended duration of administration for Herpes simplex encephalitis: at least 10 days
- Injection directly into a vein over one hour by a controlled-rate infusion pump
- Dilution of infusion solution
- Compatibility with various infusion fluids
Interaction
- Co-administration with probenecid
- Effects of diuretic use in patients over 60 years
Contraindications
- Hypersensitivity to Acyclovir
- Hypersensitivity to Valacyclovir
