Description

Dosage & Administration

  • For adults: 40000 IU once weekly for 7 weeks for Vitamin D3 deficiency
  • For adults: 1400-2000 IU/day for maintenance therapy
  • For children (12-18 years): 20000 IU once every 2 weeks for 6 weeks for Vitamin D3 deficiency
  • For film-coated tablet: 1000 IU (1-2 tablets) daily, or as directed by a physician
  • For oroflash or chewable tablets: 1000-2000 IU daily, or as directed by a physician
  • For syrup: 400 IU/day for infants (0-1 yr)
  • For syrup: 600 IU/day for children (>1 yr)
  • For injection: Various doses recommended for different age groups and conditions

Pharmacology

  • Binds to Vitamin D receptors (VDRs)
  • Has a half-life of about 50 days as it is fat-soluble
  • Absorbed in the small intestine and metabolized to Calcidiol in the liver
  • Metabolism to Calcitriol occurs in the kidney
  • Increases the absorption of calcium
  • Excreted via faeces and urine

Interaction

  • Many drugs can cause Vitamin D3 deficiencies

Contraindications

  • Not recommended in diseases associated with hypercalcemia
  • Not recommended in cases of known hypersensitivity to Vitamin D3
  • Not recommended in the presence of evidence of Vitamin D3 toxicity

Side Effects

  • Symptoms include anorexia, lassitude, nausea & vomiting, diarrhea, constipation, weight loss, polyuria, sweating, headache, thirst, vertigo, and raised concentrations of calcium and phosphate in plasma and urine

Pregnancy & Lactation

  • Safe use of doses up to 4000 IU during pregnancy
  • Recommended daily intake for pregnant women is 400 IU
  • Vitamin D3 and its metabolites are excreted in breast milk
  • No observed overdose in breastfed infants

Precautions & Warnings

  • Plasma-calcium concentration should be monitored at intervals in certain patient groups
  • Consultation required for patients using Digoxin and Thiazide Diuretics
  • Consultation required for patients with specific diseases

Use in Special Populations

  • Safety & efficacy in children under 12 years not established

Overdose Effects

  • Can lead to hypervitaminosis D
