Dosage
- 40000 IU once weekly for adults for 7 weeks
- 1000-2000 IU/day for maintenance therapy
- 20000 IU every 4 weeks for prevention in adults
- 20000 IU once a month for specific therapy for osteoporosis in adults
- 20000 IU once every 2 weeks for children (12-18 years)
- 20000 IU every 6 weeks for prevention in children (12-18 years)
- 1000 IU (1-2 tablets) daily for film-coated tablet
- 1000-2000 IU daily for oroflash or chewable tablets
- 400 IU/day (0-1 yr) for patients with risk of cholecalciferol deficiency in syrup
- 600 IU/day (>1 yr) for patients with risk of cholecalciferol deficiency in syrup
- 2000 IU/day (+50000 IU/week) for 6 weeks (0-1 yr) for cholecalciferol deficient patients in syrup
- 2000 IU/day for 6 weeks (1-18 yrs) for cholecalciferol deficient patients in syrup
- 50000-200000 IU every 6 months for infants receiving Vitamin D enriched milk in injection
- 200000 IU every 6 months for nursed infants or infants not receiving Vitamin D enriched milk or young children up to 5 years of age in injection
- 200000 IU every 6 months for adolescents in injection
- 100000 IU from the 6th or 7th month of pregnancy for pregnancy in injection
- 100000-200000 IU every 3-6 months for elderly and specific conditions in injection
Mode of Action
- Vitamin D3 is absorbed in the small intestine and metabolized to 25-hydroxy Vitamin D3 in the liver
- Calcitriol, the active form of Vitamin D3, binds to Vitamin D receptors (VDRs) in body tissues
- Calcitriol increases the absorption of calcium and phosphate

- Drugs causing Vitamin D3 deficiencies
- Drugs that interfere with the absorption and metabolism of Vitamin D3

- Hypercalcaemia
- Known hypersensitivity to Vitamin D3
- Evidence of Vitamin D3 toxicity

Precautions & Warnings
- Monitor plasma-calcium concentration in patients receiving high doses of Vitamin D3
- Consultation for people using Digoxin and Thiazide Diuretics
- Consultation for people with liver or kidney disease, primary hyperthyroidism, lymphoma, tuberculosis, and granulomatous disease
