Dosage

  • 40000 IU once weekly for adults for 7 weeks

Mode of Action

  • Vitamin D3 is absorbed in the small intestine and metabolized to 25-hydroxy Vitamin D3 in the liver
  • Calcitriol, the active form of Vitamin D3, binds to Vitamin D receptors (VDRs) in body tissues
  • Calcitriol increases the absorption of calcium and phosphate

  • Drugs causing Vitamin D3 deficiencies
  • Drugs that interfere with the absorption and metabolism of Vitamin D3

Contraindications

  • Hypercalcaemia
  • Known hypersensitivity to Vitamin D3
  • Evidence of Vitamin D3 toxicity

Precautions & Warnings

  • Monitor plasma-calcium concentration in patients receiving high doses of Vitamin D3
  • Consultation for people using Digoxin and Thiazide Diuretics
  • Consultation for people with liver or kidney disease, primary hyperthyroidism, lymphoma, tuberculosis, and granulomatous disease

Use in Special Populations

  • Safety & efficacy not established in children under 12 years
