Indications
- Post-menopausal osteoporosis
- Renal osteodystrophy
- Postsurgical hypoparathyroidism
- Idiopathic hypoparathyroidism
- Pseudohypoparathyroidism
- Secondary hyperparathyroidism in patients with moderate to severe chronic renal failure (pre-dialysis)
- Vitamin D dependent rickets
Pharmacology
- Formation in the kidneys from its precursor, 25-hydroxycholecalciferol
- Promotes intestinal absorption of calcium
- Regulates bone mineralization
- Stimulation effects on osteoblastic activity in the skeleton
Dosage & Administration
- Optimal daily dose carefully determined based on serum calcium level
- Recommended doses for different conditions
- Intravenous initial dose depending on the severity of hypocalcemia and/or secondary hyperparathyroidism
Interaction
- Increased risk of hypercalcemia with thiazide diuretics
- Careful dosage determination in patients undergoing treatment with digitalis
- Potential hypermagnesemia with magnesium containing drugs
Contraindications
- Known hypersensitivity to any of its ingredients
- All diseases associated with hypercalcemia
Side Effects
- Early and late side effects including weakness, headache, nausea, polyuria, anorexia, and elevated SGOT and SGPT
Pregnancy & Lactation
- No evidence of teratogenicity in humans
- Use during pregnancy only if benefits outweigh potential risk to the fetus
- Monitoring of serum calcium levels for breastfeeding mothers and infants
Precautions & Warnings
- Induced hypercalcemia and hypercalciuria with excessive dosage
- Caution in patients on digitalis
- Rebound effect upon discontinuation
- Potential soft-tissue calcification with chronic hypercalcemia

Overdose Effects
