Indications
- Post-menopausal osteoporosis
- Renal osteodystrophy
- Postsurgical hypoparathyroidism
- Idiopathic hypoparathyroidism
- Pseudohypoparathyroidism
- Secondary hyperparathyroidism in patients with moderate to severe chronic renal failure (pre-dialysis)
- Vitamin D dependent rickets
Pharmacology
Calcitriol promotes intestinal absorption of calcium and regulates bone mineralization. It plays a key role in the regulation of calcium homeostasis and stimulates osteoblastic activity in the skeleton.
Dosage & Administration
- Recommended daily doses for specific conditions
- Optimal daily dose must be carefully determined based on serum calcium level
Interaction
Concomitant treatment with a thiazide diuretic increases the risk of hypercalcemia. Careful dosage determination is required for patients undergoing treatment with digitalis.
Contraindications
Contraindicated in patients with known hypersensitivity to any of its ingredients and in diseases associated with hypercalcemia.
Side Effects
- Early side effects include weakness, headache, nausea, vomiting, dry mouth, muscle and bone pain, metallic taste.
- Late side effects include polyuria, anorexia, weight loss, nocturia, conjunctivitis, pancreatitis, hyperthermia, hypertension, and more.
Pregnancy & Lactation
May be used during pregnancy if benefits outweigh potential risks. Breastfeeding mothers should monitor serum calcium levels of both the mother and infant
Precautions & Warnings
- Excessive dosage may induce hypercalcemia and hypercalciuria
- Caution for patients on digitalis, discontinuation may result in rebound effect
- The most potent metabolite of vitamin D, Vitamin D and its derivatives should be withheld during treatment

Overdose Effects
Can cause hypercalcemia, hypercalciuria, and hyperphosphatemia. General treatment includes immediate discontinuation, low calcium diet, and withdrawal of calcium supplements.
