Indications
- Treatment of thiamine deficiency such as Beriberi and Wernick's encephalopathy
- Neuritis associated with pregnancy
- Pellagra
Pharmacology
- Coenzyme for decarboxylation of α-ketoglutaric acid
- Affects the peripheral nervous system, gastrointestinal tract, and cardiovascular system
- Necessary for optimal growth of infants and children
- Not stored in the body and regularly lost during deficiency
- Elevated requirements during pregnancy, carbohydrate rich diet, and alcoholism
- Effects of thiamine deficiency: fatigue, anorexia, gastrointestinal disturbance, tachycardia, irritability, neurological symptoms
- Well absorption from the gastrointestinal tract
- Rapid absorption from the upper small intestine
- Not stored in the body to a significant extent
- Plasma half life of 24 hours
Dosage & Administration
- Prophylaxis: 3 to 10 mg daily
- Mild chronic deficiency: 10 to 25 mg daily
- Severe deficiency: 200 to 300 mg daily

- No hazardous drug interactions reported
- Acts synergistically with other B-complex group vitamins
Contraindications
- No absolute contraindication
- Risk of anaphylaxis increased by repeated parenteral administration
- Mild allergic phenomena may lead to anaphylactic shock
- Injection should not be given intravenously except in comatose patients

- No adverse effects when given orally
- Anaphylactic reactions with large intravenous doses in sensitive patients
- Risk of reactions increases with repeated parenteral administration
- Transient mild soreness at the site of intramuscular administration
