Description

A dopamine antagonist medication used to treat dyspeptic symptom complex, acute nausea and vomiting, Parkinson's disease, and speeding barium transit in follow-through radiological studies.

DosageInstructions

  • Adults:
    • 10-20 mg (1-2 tablet or 10-20 ml suspension), every 6-8 hours daily
    • 30-60 mg rectally every 4-8 hours
  • Children:
    • 2-4 ml suspension/10 kg body weight or 0.4-0.8 ml pediatric drops/10 kg body weight, every 6-8 hours daily
    • 0.2-0.4 mg/kg body weight, every 6-8 hours daily

Pharmacology

Dopamine antagonist that blocks receptors in Chemoreceptor Trigger Zone and stomach, restores normal motility and tone of the upper gastrointestinal tract, enhances gastric emptying, regulates contraction of the pylorus, and increases esophageal peristalsis.

Interaction

  • Antagonized by anticholinergic drugs
  • Lower oral bioavailability with antacids and antisecretory drugs
  • Metabolic pathway through CYP3A4
  • Potential influence on absorption of concomitantly orally administered drugs

Contraindications

  • Known hypersensitivity
  • Neonates
  • Gastrointestinal hemorrhage, mechanical obstruction, perforation
  • Prolactin releasing pituitary tumor (prolactinoma)
