Indications
- Metastatic Testicular Tumors
- Metastatic Ovarian Tumors
- Advanced Bladder Cancer
- Small Cell Lung Carcinoma
Pharmacology
- Platinum compound
- Inhibits DNA synthesis
- Inhibits protein and RNA synthesis
- Produces intra-and interstrand cross links
Dosage & Administration
- Administered by slow intravenous infusion
- Should not be given by rapid intravenous injection
- Usual dose for testicular cancer: 20 mg/m2 IV daily for 5 days per cycle
- Usual dose for metastatic ovarian tumors: 75 to 100 mg/m2 IV per cycle
Interaction
- Additive toxicity with antineoplastic drugs
- Nephrotoxicity with aminoglycoside antibiotics
- Ototoxicity with aminoglycoside antibiotics and loop diuretics
- Alters renal elimination of certain drugs
Contraindications
- Pre-existing renal impairment
- History of hypersensitivity to Cisplatin
- Myelosuppresed patients
- Patients with hearing impairment
Side Effects
- Severe nausea and vomiting
- Electrolyte disturbances: hypomagnesemia, hypocalcemia, hypokalemia
- Renal tubular dysfunction
