Description
- Synthetic purine derivative
- Treats Herpes simplex virus (HSV) and Varicella-zoster virus infections
- Interferes with DNA synthesis and inhibits viral replication
- Administered with caution in patients with renal impairment
- Pregnancy category B
- Should be stored below 25°C and protected from light and moisture
- Probenecid reduces Aciclovir excretion and increases plasma concentration and risk of toxicity
Indications
- Treatment of viral infections due to Herpes simplex virus (type I & II) and Varicella zoster virus (herpes zoster & chicken pox)
- Treatment of Herpes simplex virus infections of the skin and mucous membranes including initial and recurrent genital herpes and herpes labialis
- Prophylaxis of Herpes simplex infections in immunocompromised patients
Mode of action
- Exerts antiviral effect by interfering with DNA synthesis and inhibiting viral replication
- Converted to Aciclovir Triphosphate, the pharmacologically active form of the drug
- 15-30% of an oral dose is absorbed from the gastrointestinal tract
- Widely distributed into body tissues and fluids
- Excreted through the kidney by glomerular filtration & tubular secretion
Dosage and administration
- 200 mg 5 times daily for 5 days for initial herpes simplex treatment
- 400 mg 5 times daily for 5 days for immunocompromised patients
- 800 mg 5 times daily for genital herpes in immunocompromised patients
- 200 mg 4 times daily for prevention of recurrence of herpes simplex
- 200 to 400 mg 4 times daily for prophylactic treatment of herpes simplex in immunocompromised patients
- 800 mg 4 times daily for 5 days for chicken pox in adults and children over 40kg
- 800 mg 5 times daily for 7 days for herpes zoster (shingles) treatment
- 400 mg 5 times daily for 10 days for initial rectal (proctitis) herpes infections
- Dose reduction recommended for patients with severe renal impairment

- Probenecid reduces Aciclovir excretion and increases plasma concentration and risk of toxicity

- Hypersensitivity to Aciclovir
