Description
- Taxotere is a microtubule inhibitor indicated for various types of cancer including Breast Cancer (BC), Non-small Cell Lung Cancer (NSCLC), Castration-Resistant Prostate Cancer (CRPC), Gastric Adenocarcinoma (GC), and Squamous Cell Carcinoma of the Head and Neck (SCCHN). It acts by disrupting the microtubular network in cells that is essential for vital mitotic and interphase cellular functions.
Dosage and administration
- Administered intravenously over 1 hr every 3 weeks
- Dosage varies based on the type of cancer being treated
Pharmacology
- Acts by disrupting the microtubular network in cells
- Promotes the assembly of tubulin into stable microtubules while inhibiting their disassembly
- Binds to free tubulin, decreasing the critical intracellular concentration of tubulin
Interaction
- Taxotere is a CYP3A4 substrate
- Metabolism may be modified by concomitant administration of other compounds
Contraindications
- Contraindicated in patients with neutrophil counts of <1500 cells/mm3
- Contraindicated in patients with a history of severe hypersensitivity reactions to docetaxel or to other drugs formulated with polysorbate 80
Side effects
- Common adverse reactions include infections, neutropenia, anemia, febrile neutropenia, hypersensitivity, thrombocytopenia, neuropathy, dysgeusia, dyspnea, constipation, anorexia, nail disorders, fluid retention, asthenia, pain, and more
Pregnancy and lactation
- Can cause fetal harm when administered to a pregnant woman
- No information regarding presence in human milk or effects on breastfed child
Precautions and warnings
- Should be administered under the supervision of a physician experienced in the use of antineoplastic agents
- Higher risk of severe adverse reactions in patients with hepatic impairment
Overdose effects
- No known antidote for docetaxel overdose
- Expected complications include bone marrow suppression, peripheral neurotoxicity, and mucositis
