Pharmacology
- Disrupts microtubular network in cells
- Promotes the assembly of tubulin into stable microtubules
- Inhibits disassembly of microtubules
- Binds to free tubulin
- Decreases critical intracellular concentration of tubulin
Dosage & Administration
- Administered intravenously (IV) over 1 hr every 3 weeks
- Dosage ranges for different cancer types
- Facility equipped to manage complications needed for administration
- Use of PVC equipment not recommended
- Premedication with oral corticosteroids required
- Dose adjustment as needed
Interaction
- CYP3A4 substrate
- Metabolism may be modified by concomitant administration of certain compounds
Contraindications
- Patients with neutrophil counts of <1500 cells/mm3
- History of severe hypersensitivity reactions to docetaxel
- History of severe hypersensitivity reactions to drugs formulated with polysorbate 80
