Indications
- Treatment of newly diagnosed glioblastoma multiforme
- Treatment of refractory anaplastic astrocytoma
Pharmacology
- Undergoes rapid nonenzymatic conversion at physiologic pH
- Reactive compound Methyl Triazen Imidazole Carboxamide (MTIC)
- Cytotoxicity due to alkylation of DNA at guanine positions
Dosage
- Newly Diagnosed GBM: 75 mg/m2 for 42 days concomitant with radiotherapy
- Newly Diagnosed GBM: Maintenance dose of 150 mg/m2 once daily
- Refractory Anaplastic Astrocytoma: Initial dose 150 mg/m2 once daily
Administration
- Should be taken on an empty stomach
- Take at least 1 hr before meals
Interaction
- Administration of valproic acid decreases oral clearance by about 5%
Contraindications
- History of hypersensitivity reaction to any of its components
- History of hypersensitivity to DTIC

- Myelosuppression monitoring required
- Pneumocystis carinii pneumonia (PCP) prophylaxis required
- Fetal harm potential
- Avoid becoming pregnant while receiving Zolomide
Use in Special Populations
- Safety and effectiveness in pediatric patients not established
- Caution with severe renal impairment
- Caution with severe hepatic impairment
Overdose Effects
- Dose-limiting toxicity is hematologic
- Adverse reactions reported include bone marrow suppression and infections
