Indications
- Newly Diagnosed Glioblastoma Multiforme
- Refractory Anaplastic Astrocytoma
Pharmacology
- Nonenzymatic conversion of Temozolomide to reactive compound MTIC
- DNA alkylation at guanine positions
Dosage
- Dosage for Newly Diagnosed GBM: 75 mg/m² for 42 days concomitant with radiotherapy
- Maintenance dose: 150 mg/m² once daily for 6 cycles
- Dosage for Refractory Anaplastic Astrocytoma: Initial dose 150 mg/m² once daily for 5 consecutive days per 28-day treatment cycle
Administration
- Take on an empty stomach
- Take at least 1 hr before meals
Interaction
- Decrease in oral clearance with Valproic Acid
Contraindications
- Hypersensitivity to any of its components
- Hypersensitivity to DTIC
