Usability Features

Dosage & Administration

  • Should be taken 15-30 minutes before meals
  • Adults: 10-20 mg every 6-8 hours daily, Children: 2-4 ml suspension/10 kg body weight every 6-8 hours daily
  • Maximum period of treatment is 12 weeks

Pharmacology

Interaction

  • Concomitant administration of anticholinergic drugs may antagonize the antidyspeptic effect of domperidone
  • Antacids and antisecretory drugs should not be given simultaneously with domperidone
  • In vitro data suggest that concomitant use of drugs inhibiting CYP3A4 may result in increased plasma levels of domperidone

Contraindications

  • Known hypersensitivity to domperidone
  • Neonates
  • Gastrointestinal hemorrhage, mechanical obstruction, or perforation
  • Patients with prolactin-releasing pituitary tumor

Side Effects

  • Rare cases of transient intestinal cramps
  • Extrapyramidal phenomena rare in young children and exceptional in adults
  • Possible increase in plasma prolactin level
  • Rare allergic reactions such as rash and urticaria

Pregnancy & Lactation

  • Should only be used during the first trimester of pregnancy if justified by therapeutic benefit
  • Not recommended during nursing unless expected benefits outweigh potential risk

  • Use with caution in children due to the risk of extra-pyramidal reactions
  • Use with caution in patients with hepatic impairment
