Description

Usability Features

Dosage & Administration
- Should be taken 15-30 minutes before meals
- Adults: 10-20 mg every 6-8 hours daily, Children: 2-4 ml suspension/10 kg body weight every 6-8 hours daily
- Maximum period of treatment is 12 weeks
Pharmacology
- Dopamine antagonist
- Blocks dopamine receptors in the Chemoreceptor Trigger Zone (CTZ) and stomach
- Weak penetration across the blood-brain barrier
- No psychotropic and neurologic side effects

Interaction
- Concomitant administration of anticholinergic drugs may antagonize the antidyspeptic effect of domperidone
- Antacids and antisecretory drugs should not be given simultaneously with domperidone
- In vitro data suggest that concomitant use of drugs inhibiting CYP3A4 may result in increased plasma levels of domperidone
Contraindications
- Known hypersensitivity to domperidone
- Neonates
- Gastrointestinal hemorrhage, mechanical obstruction, or perforation
- Patients with prolactin-releasing pituitary tumor
Side Effects
- Rare cases of transient intestinal cramps
- Extrapyramidal phenomena rare in young children and exceptional in adults
- Possible increase in plasma prolactin level
- Rare allergic reactions such as rash and urticaria
Pregnancy & Lactation
- Should only be used during the first trimester of pregnancy if justified by therapeutic benefit
- Not recommended during nursing unless expected benefits outweigh potential risk
Precautions & Warnings
- Use with caution in children due to the risk of extra-pyramidal reactions
- Use with caution in patients with hepatic impairment
