Usability Features

Dosage & Administration

  • 10-20 mg (1-2 tablet or 10-20 ml suspension) every 6-8 hours daily for adults
  • 2-4 ml suspension/10 kg body weight or 0.4-0.8 ml paediatric drops/10 kg body weight every 6-8 hours daily for children

Drug Interactions

  • Anticholinergic drugs may antagonize the antidyspeptic effect of domperidone
  • Antacids and antisecretory drugs lower oral bioavailability of domperidone
  • Main metabolic pathway of domperidone is through CYP3A4
  • In vitro data suggest that concomitant use of drugs that inhibits CYP3A4 may result in increased plasma levels of domperidone

Contraindications

  • Known hypersensitivity to domperidone
  • Neonates
  • Gastrointestinal hemorrhage, mechanical obstruction, or perforation
  • Prolactin releasing pituitary tumor (prolactinoma)

Pregnancy & Lactation

  • Should only be used during the first trimester of pregnancy if justified by anticipated therapeutic benefit
  • Not recommended for nursing mothers unless expected benefits outweigh potential risk
  • Does not affect mental alertness

Precautions & Warnings

  • Increased risk of extra-pyramidal reactions in young children
  • Should be used with caution in patients with hepatic impairment

Overdose Effects

  • Symptoms may include drowsiness, disorientation, and extrapyramidal reactions
  • Administration of activated charcoal and close observation recommended
