Indications
- Short-term management of moderate to severe acute post-operative pain
- Relief of pain, inflammation, and light sensitivity after ocular surgery
- Decreasing eye inflammation or redness caused by seasonal allergies
Description
- Belongs to pyrrolo-pyrrole group of nonsteroidal anti-inflammatory drug (NSAID)
- Chemical name: 5 benzoyle-2,3-dihydro-1H-pyrroligine-1-carboxylic acid, compound with 2-amino-2-(hydroxymethyl)-1,3-propanediol
- Inhibits synthesis of prostaglandins
- Biological activity associated with S-form
- Pharmacokinetic properties: Linear, highly protein bound, mostly metabolized in liver
Pharmacology
- Potent analgesic of the non-steroidal anti-inflammatory drugs (NSAIDs)
- Inhibits cyclooxygenase enzyme system and prostaglandin synthesis
- Minimal anti-inflammatory effect at analgesic dose
Dosage & Administration
- Tablet: Recommended dose of 10 mg every 4-6 hours for short-term use (up to 7 days)
- Injection: Single or multiple doses for moderately severe, acute pain management, with specific recommended doses based on age, renal function, and body weight
- Eye Drops: Recommended dosage of 1 drop in each eye 4 times daily
Interaction
- Other NSAIDs or aspirin: Increases side effects
- Anti-coagulants: Enhances anti-coagulant effect
- Beta Blocker: Reduces anti-hypertensive effect
- ACE Inhibitors: Increases risk of renal impairment
- Methotrexate: Enhances toxicity of methotrexate
Contraindications
- Hypersensitivity to Ketorolac or other NSAIDs
- Children under 16 years of age
- Prophylactic analgesic before surgery

- Nausea, vomiting, gastro-intestinal bleeding, melena, peptic ulcer, pancreatitis, anxiety, drowsiness, headache, excessive thirst, fatigue, bradycardia, hypertension, palpitation, chest pain, infertility in females, pulmonary edema

- Avoid in pregnancy and lactation unless benefits outweigh risks to fetus
Precautions & Warnings
- Caution in patients over 65 years
- Caution in patients with active or suspected peptic ulcer, gastrointestinal bleeding, asthma, and liver dysfunction
- Eye Drops: Discontinue use with evidence of corneal epithelial breakdown, avoid while wearing contact lenses
