Indications
- Short-term management of moderate to severe post-operative pain
- Relief of pain, inflammation, and light sensitivity after ocular surgery

Description
- Belongs to pyrrolo-pyrrole group of nonsteroidal anti-inflammatory drug (NSAID)
- Chemical composition: 5 benzoyle-2,3-dihydro-1H-pyrroligine-1-carboxylic acid, compound with 2-amino-2-(hydroxymethyl)-1,3-propanediol (1:1)
- Inhibits prostaglandin synthesis
- Biological activity associated with the S-form
- Linear pharmacokinetic property
- Highly protein bound
- Metabolized in the liver
- Metabolites and some unchanged drugs excreted in urine
Pharmacology
- Potent analgesic of the non-steroidal anti-inflammatory drugs (NSAIDs)
- Inhibits cyclooxygenase enzyme system
- Minimal anti-inflammatory effect at analgesic dose
Dosage & Administration
- Recommended tablet dose: 10 mg every 4-6 hours for up to 7 days, not for chronic use
- Recommended injection dose: may be used as single or multiple doses for moderately severe, acute pain that requires analgesia at the opioid level
- Onset of analgesic effect within 30 minutes for IV or IM administration
- Duration of analgesic effect usually 4 to 6 hours
- Specific dosing instructions for different patient groups
Interaction
- Increased side effects with other NSAIDs or aspirin
- Enhanced anti-coagulant effect with anti-coagulants
- Reduced anti-hypertensive effect with beta blockers
- Increased risk of renal impairment with ACE inhibitors
- Enhanced toxicity of methotrexate
Contraindications
- Hypersensitivity to Ketorolac or other NSAIDs
- Children under 16 years of age
- Prophylactic analgesic before surgery

- Discontinue use of topical NSAIDs in patients with evidence of corneal epithelial breakdown
- Avoid administering while wearing contact lenses
