Indications
- Short-term management of moderate to severe acute post-operative pain

Description
- Pyrrolo-pyrrole group of nonsteroidal anti-inflammatory drug (NSAID)
- Chemical composition: 5 benzoyle-2,3-dihydro-1H-pyrroligine-1-carboxylic acid
- Inhibited synthesis of prostaglandins
- Biological activity associated with the S-form
- Linear pharmacokinetic property
- Highly protein bound
- Largely metabolized in the liver
- Metabolites and unchanged drugs excreted in urine
Pharmacology
- Potent analgesic of the non-steroidal anti-inflammatory drugs (NSAIDs)
- Inhibits cyclooxygenase enzyme system
- Minimal anti-inflammatory effect at analgesic dose
Dosage & Administration
- Tablet recommended dose: 10 mg every 4-6 hours, not for chronic use
- Injection for postoperative pain management
- Different doses for patients based on age, weight, and renal function
- Recommended dosage for eye drops: 1 drop in each eye 4 times daily
Interaction
- Enhanced side effects with other NSAIDs or aspirin
- Enhanced anti-coagulant effect with anti-coagulants
- Reduced anti-hypertensive effect with beta blockers
- Increased risk of renal impairment with ACE inhibitors
- Enhanced methotrexate toxicity
Contraindications
- Hypersensitivity to ketorolac or other NSAIDs
- Children under 16 years of age
- Prophylactic analgesic use before surgery
