Indications
- Short-term management of post-operative pain

Description
- Belongs to pyrrolo-pyrrole group of nonsteroidal anti-inflammatory drug (NSAID)
- Chemical name: 5 benzoyle-2,3-dihydro-1H-pyrroligine-1-carboxylic acid, compound with 2 - amino-2-(hydroxymethyl)-1,3-propanediol (1:1)
- Inhibits prostaglandin synthesis and acts as a peripherally acting analgesic
- Biological activity associated with the S-form
- Linear pharmacokinetic property
- Highly protein bound and largely metabolized in the liver
Pharmacology
- Potent analgesic of the non-steroidal anti-inflammatory drugs (NSAIDs)
- Inhibits the cyclooxygenase enzyme system and prostaglandin synthesis
- Minimal anti-inflammatory effect at analgesic dose
Dosage & Administration
- Tablet recommended dose: 10 mg every 4-6 hours, up to 7 days only
- Injection used for moderately severe, acute pain on regular or as-needed schedule
- IV bolus administration over at least 15 seconds
- IM administration given slowly and deeply into the muscle
- Analgesic effect begins within 30 minutes, with maximum effect in 1 to 2 hours after dosing IV or IM
- Duration of analgesic effect is usually 4 to 6 hours
- Specific dosing guidelines for different age groups and conditions
Interaction
- Other NSAIDs or aspirin increase side effects
- Anti-coagulants enhance anti-coagulant effect
- Beta Blockers reduce anti-hypertensive effect
- ACE Inhibitors increase risk of renal impairment
- Methotrexate enhances toxicity of methotrexate
Contraindications
- Hypersensitivity to Ketorolac or other NSAIDs
- Children under 16 years of age
- Prophylactic analgesic use before surgery

- Eye Drops: Discontinue use if evidence of corneal epithelial breakdown, avoid use with contact lenses
