Indications
- Short-term management of moderate to severe acute post-operative pain
- Relieves pain, inflammation, and light sensitivity after ocular surgery
- Decreases eye inflammation or redness caused by seasonal allergies
Description
- Belongs to the pyrrolo-pyrrole group of nonsteroidal anti-inflammatory drug (NSAID)
- Chemical name is 5 benzoyle-2,3-dihydro-1H-pyrroligine-1-carboxylic acid, compound with 2 - amino-2-(hydroxymethyl)-1,3-propanediol (1:1)
- Inhibits synthesis of prostaglandins
- Biological activity associated with the S-form
- Highly protein bound
- Metabolized in the liver
Pharmacology
- Potent analgesic of the non-steroidal anti-inflammatory drugs (NSAIDs)
- Inhibits the cyclooxygenase enzyme system
- Demonstrates minimal anti-inflammatory effect at its analgesic dose
Dosage & Administration
- Tablet recommended dose of 10 mg every 4-6 hours for short-term use
- Tablet not recommended for chronic use or doses exceeding 40 mg/day
- Injection may be used for moderately severe, acute pain
- IV bolus must be given over no less than 15 seconds
- Maximum analgesic effect seen within 1 to 2 hours after dosing
- Duration of analgesic effect is usually 4 to 6 hours
- Eye drops recommended 1 drop in each eye 4 times daily
Interaction
- Other NSAIDs or aspirin can increase side effects
- Anti-coagulants can enhance anti-coagulant effect
- Beta Blockers can reduce the anti-hypertensive effect
- ACE Inhibitors can increase the risk of renal impairment
- Methotrexate can enhance the toxicity of methotrexate
Contraindications
- Hypersensitivity to Ketorolac or other NSAIDs
- Not to be used in children under 16 years of age
- Contraindicated as prophylactic analgesic before surgery

Pregnancy & Lactation

- Ketorolac Tromethamine should be avoided in pregnancy and lactation unless the potential benefits outweigh the risks to the fetus
Precautions & Warnings
- Caution in patients over the age of 65 years
- Caution in patients with active or suspected peptic ulcer, gastrointestinal bleeding, asthma, and liver dysfunction
- Topical NSAIDs should be discontinued in patients with evidence of corneal epithelial breakdown
- Eye drops should not be administered while wearing contact lenses
