Indications
- Short-term management of moderate to severe acute post-operative pain
- Relief of pain, inflammation, and light sensitivity after ocular surgery
- Decrease in eye inflammation or redness caused by seasonal allergies
Description
- Belongs to the pyrrolo-pyrrole group of nonsteroidal anti-inflammatory drugs (NSAID)
- Chemical composition as 5 benzoyle-2,3-dihydro-1H-pyrroligine-1-carboxylic acid, compound with 2 - amino-2-(hydroxymethyl)-1,3-propanediol (1:1)
- Inhibits synthesis of prostaglandins and considered as a peripherally acting analgesic
- Biological activity associated with the S-form
- Pharmacokinetic property is linear
- Highly protein bound and largely metabolized in the liver
- Metabolites and some unchanged drugs are excreted in the urine
Pharmacology
- Potent analgesic of the non-steroidal anti-inflammatory drugs (NSAIDs)
- Inhibits the cyclooxygenase enzyme system and prostaglandin synthesis
- Minimal anti-inflammatory effect at analgesic dose
Dosage & Administration
- Recommended dose of 10 mg tablet every 4-6 hours for short-term use (up to 7 days)
- Recommended dosage for ketorolac injection in different patient groups and age brackets
- Recommended dosage for eye drops: Adults - 1 drop in each eye 4 times daily
Interaction
- Interaction with other NSAIDs or aspirin
- Interaction with anti-coagulants, beta blockers, and ACE inhibitors
- Enhanced toxicity of methotrexate
Contraindications
- Hypersensitivity to Ketorolac or other NSAIDs
- Use in children under 16 years of age
- Prophylactic analgesic use before surgery

- Nausea, vomiting, and gastrointestinal bleeding
- Melana, peptic ulcer, and pancreatitis
- Anxiety, drowsiness, and headache
- Excessive thirst, fatigue, and bradycardia
- Hypertension, palpitation, and chest pain
- Infertility in females and pulmonary edema

Precautions & Warnings
- Caution in patients over the age of 65 years
