Indications

  • Short-term management of moderate to severe acute post-operative pain
  • Relief of pain, inflammation, and light sensitivity after ocular surgery
  • Decrease in eye inflammation or redness caused by seasonal allergies

Description

  • Belongs to the pyrrolo-pyrrole group of nonsteroidal anti-inflammatory drugs (NSAID)
  • Chemical composition as 5 benzoyle-2,3-dihydro-1H-pyrroligine-1-carboxylic acid, compound with 2 - amino-2-(hydroxymethyl)-1,3-propanediol (1:1)
  • Inhibits synthesis of prostaglandins and considered as a peripherally acting analgesic
  • Biological activity associated with the S-form
  • Pharmacokinetic property is linear
  • Highly protein bound and largely metabolized in the liver
  • Metabolites and some unchanged drugs are excreted in the urine

Pharmacology

  • Potent analgesic of the non-steroidal anti-inflammatory drugs (NSAIDs)
  • Inhibits the cyclooxygenase enzyme system and prostaglandin synthesis
  • Minimal anti-inflammatory effect at analgesic dose

Dosage & Administration

  • Recommended dose of 10 mg tablet every 4-6 hours for short-term use (up to 7 days)
  • Recommended dosage for ketorolac injection in different patient groups and age brackets
  • Recommended dosage for eye drops: Adults - 1 drop in each eye 4 times daily

Interaction

  • Interaction with other NSAIDs or aspirin
  • Interaction with anti-coagulants, beta blockers, and ACE inhibitors
  • Enhanced toxicity of methotrexate

Contraindications

  • Hypersensitivity to Ketorolac or other NSAIDs
  • Use in children under 16 years of age
  • Prophylactic analgesic use before surgery

  • Nausea, vomiting, and gastrointestinal bleeding
  • Melana, peptic ulcer, and pancreatitis
  • Anxiety, drowsiness, and headache
  • Excessive thirst, fatigue, and bradycardia
  • Hypertension, palpitation, and chest pain

Precautions & Warnings

  • Caution in patients over the age of 65 years

  • Special precautions for patients using eye drops and wearing contact lenses
