Description

  • Belongs to the pyrrolo-pyrrole group of NSAIDs
  • Chemical composition: 5 benzoyle-2,3-dihydro-1H-pyrroligine-1-carboxylic acid, compound with 2-amino-2-(hydroxymethyl)-1,3-propanediol (1:1)
  • Inhibits synthesis of prostaglandins
  • Biologically active form is the S-form
  • Pharmacokinetic property is linear
  • Highly protein bound
  • Largely metabolized in the liver
  • Excretion in the urine

Pharmacology

  • Acts by inhibiting the cyclooxygenase enzyme system
  • Minimal anti-inflammatory effect at its analgesic dose

Dosage & Administration

  • Tablet recommended dose: 10 mg every 4-6 hours for up to 7 days
  • Injection for postoperative pain management
  • Onset of analgesic effect within 30 minutes
  • Maximum effect in 1 to 2 hours after dosing IV or IM
  • Duration of analgesic effect: 4 to 6 hours
  • Specific dosages for different age groups and conditions

Interactions

  • Increased side effects with other NSAIDs or aspirin
  • Enhanced anti-coagulant effect with anti-coagulants
  • Reduced anti-hypertensive effect with Beta Blockers
  • Increased risk of renal impairment with ACE Inhibitors

Contraindications

  • Hypersensitivity to Ketorolac or other NSAIDs
