Indications

  • Short-term management of moderate to severe acute post-operative pain
  • Relieves pain, inflammation, and light sensitivity after ocular surgery
  • Decreases eye inflammation or redness caused by seasonal allergies

Description

  • Belongs to pyrrolo-pyrrole group of nonsteroidal anti-inflammatory drug (NSAID)
  • Chemical composition: 5 benzoyle-2,3-dihydro-1H-pyrroligine-1-carboxylic acid, compound with 2 - amino-2-(hydroxymethyl)-1,3-propanediol (1:1)
  • Inhibits synthesis of prostaglandins
  • Peripherally acting analgesic
  • Biologically active form: S-form
  • Linear pharmacokinetic property
  • Highly protein bound
  • Metabolized in the liver
  • Excretion in urine

Pharmacology

  • Potent analgesic
  • Inhibits cyclooxygenase enzyme system
  • Minimal anti-inflammatory effect at analgesic dose

Dosage & Administration

  • Tablet dosage: 10 mg every 4-6 hours for up to 7 days
  • Injection for acute pain management
  • IV bolus administration time: not less than 15 seconds
  • Onset of analgesic effect within 30 minutes to 1-2 hours
  • Duration of analgesic effect: 4 to 6 hours
  • Single and multiple dose treatment guidelines for different patient populations
  • Recommended dosage for ophthalmic solution

Interaction

  • Increased side effects with other NSAIDs or aspirin
  • Enhanced anti-coagulant effect with anti-coagulants
  • Reduction of anti-hypertensive effect with Beta Blocker
  • Increased risk of renal impairment with ACE Inhibitors
  • Enhanced toxicity of methotrexate

Contraindications

  • Hypersensitivity to Ketorolac or other NSAIDs
  • Use not recommended for children under 16 years of age
  • Contradicted as prophylactic analgesic before surgery
