Indications

Description

belongs to pyrrolo-pyrrole group of nonsteroidal anti-inflammatory drug (NSAID)
chemical name: 5 benzoyle-2,3-dihydro-1H-pyrroligine-1-carboxylic acid, compound with 2 - amino-2-(hydroxymethyl)-1,3-propanediol (1:1)
inhibits synthesis of prostaglandins
S-form is associated with the biological activity
highly protein bound
largely metabolized in liver
metabolites and some unchanged drugs excreted in urine

Pharmacology

potent analgesic
inhibits cyclooxygenase enzyme system
minimal anti-inflammatory effect at analgesic dose

Dosage & Administration

10 mg tablet every 4-6 hours for up to 7 days
Injection: single or multiple doses, regular or when necessary schedule
IV bolus to be given over no less than 15 seconds
analgesic effect begins within 30 minutes
maximum effect in 1 to 2 hours after dosing IV or IM
duration of analgesic effect usually 4 to 6 hours
specific dosages for different age groups and conditions

Interaction

Other NSAIDs or aspirin: Increase side effects of Kflam
Anti-coagulants: Enhance anti-coagulant effect
Beta Blocker: Reduce anti-hypertensive effect
ACE Inhibitors: Increase risk of renal impairment
Methotrexate: Enhance toxicity of methotrexate

Contraindications

hypersensitivity to Ketorolac or other NSAIDs
not to be used in children under 16 years of age
contraindicated as prophylactic analgesic before surgery
