Indications
- short-term management of moderate to severe acute post-operative pain
- relief of pain, inflammation, and light sensitivity after ocular surgery
- decreasing eye inflammation or redness caused by seasonal allergies
Description
- drug of pyrrolo-pyrrole group of nonsteroidal anti-inflammatory drug (NSAID)
- chemical composition: 5 benzoyle-2,3-dihydro-1H-pyrroligine-1-carboxylic acid, compound with 2-amino-2-(hydroxymethyl)-1,3-propanediol (1:1)
- inhibition of prostaglandin synthesis
- peripherally acting analgesic
- metabolized in the liver
- excretion in the urine
Pharmacology
- potent analgesic
- inhibition of cyclooxygenase enzyme system
- minimal anti-inflammatory effect at analgesic dose
Dosage & Administration
- tablet recommended dose: 10 mg every 4-6 hours for up to 7 days
- contraindicated for chronic use
- injection for moderately severe, acute pain management
- IV bolus administration over at least 15 seconds
- onset of analgesic effect within 30 minutes
- maximum effect in 1 to 2 hours
- duration of analgesic effect: 4 to 6 hours
- multiple dosing based on age and renal function
- eye drops dosage: 1 drop in each eye 4 times daily
Interaction
- increase in side effects with other NSAIDs or aspirin
- enhanced anti-coagulant effect with anti-coagulants
- reduced anti-hypertensive effect with beta blockers
- increased risk of renal impairment with ACE inhibitors
- enhanced toxicity of methotrexate
Contraindications
- hypersensitivity to ketorolac or other NSAIDs
- age under 16 years
- prophylactic analgesic before surgery
