Indications

  • Short-term management of moderate to severe acute post-operative pain
  • Relief of pain, inflammation, and light sensitivity after ocular surgery
  • Decrease eye inflammation or redness caused by seasonal allergies

Description

  • Belongs to the pyrrolo-pyrrole group of nonsteroidal anti-inflammatory drugs (NSAID)
  • Chemical composition known as 5 benzoyle-2,3-dihydro-1H-pyrroligine-1-carboxylic acid, compound with 2 - amino-2-(hydroxymethyl)-1,3-propanediol (1:1)
  • Inhibits synthesis of prostaglandins
  • Biological activity associated with the S-form
  • Highly protein bound
  • Metabolized in the liver
  • Products of metabolism and some unchanged drugs are excreted in the urine
  • Linear pharmacokinetic property

Pharmacology

  • Potent analgesic of the non-steroidal anti-inflammatory drugs (NSAIDs)
  • Acts by inhibiting the cyclooxygenase enzyme system
  • Minimal anti-inflammatory effect at analgesic dose

Dosage & Administration

  • Tablet dosage: Recommended dose is 10 mg every 4-6 hours for short-term use
  • Injection dosage: Single or multiple doses for the management of moderately severe, acute pain postoperatively
  • Eye Drops dosage: 1 drop in each eye 4 times daily

Interaction

  • Other NSAIDs or aspirin increase the side effects of Knil
  • Anti-coagulants enhance anti-coagulant effect
  • Beta Blockers reduce the anti-hypertensive effect
  • ACE Inhibitors increase the risk of renal impairment
  • Methotrexate enhances the toxicity of methotrexate

Contraindications

  • Hypersensitivity to Ketorolac or other NSAIDs
  • Contraindicated in children under 16 years of age
  • Contraindicated as prophylactic analgesic before surgery

Precautions & Warnings

  • Caution in patients over the age of 65 years
  • Caution in patients with active or suspected peptic ulcer or gastrointestinal bleeding
  • Caution in patients with asthma and liver dysfunction
  • Discontinue use of topical NSAIDs if evidence of corneal epithelial breakdown
  • Do not administer while wearing contact lenses
