Indications

  • Short-term management of moderate to severe acute post-operative pain
  • Relieves pain, inflammation, and light sensitivity after ocular surgery
  • Decreases eye inflammation or redness caused by seasonal allergies

Description

  • Belongs to pyrrolo-pyrrole group of nonsteroidal anti-inflammatory drug (NSAID)
  • Inhibits synthesis of prostaglandins
  • Peripherally acting analgesic
  • Linear pharmacokinetic property
  • Highly protein bound
  • Metabolized in the liver
  • Metabolites and some unchanged drugs excreted in urine

Pharmacology

  • Potent analgesic NSAID
  • Inhibits cyclooxygenase enzyme system
  • Minimal anti-inflammatory effect at analgesic dose

Dosage & Administration

  • Tablet recommended dose of 10 mg every 4-6 hours for short-term use only, not chronic use
  • Injection may be used as single or multiple doses for moderately severe acute pain requiring analgesia at the opioid level
  • Eye drops recommended dose of 1 drop in each eye 4 times daily

Interaction

  • Other NSAIDs or aspirin increase side effects of Lacor
  • Anti-coagulants enhance anti-coagulant effect
  • Beta blocker reduces anti-hypertensive effect
  • ACE inhibitors increase risk of renal impairment
  • Methotrexate enhances toxicity of methotrexate

Contraindications

  • Hypersensitivity to Ketorolac or other NSAIDs
  • Not to be used in children under 16 years of age
  • Contraindicated as prophylactic analgesic before surgery

Precautions & Warnings

  • Caution in patients over 65 years
  • Caution in patients with active or suspected peptic ulcer, gastrointestinal bleeding, asthma, and liver dysfunction
  • Discontinue use of topical NSAIDs in patients with evidence of corneal epithelial breakdown
  • Do not administer while wearing contact lenses
