Indications

Description

  • Belongs to pyrrolo-pyrrole group of nonsteroidal anti-inflammatory drugs (NSAID)
  • Chemical name: 5 benzoyle-2,3-dihydro-1H-pyrroligine-1-carboxylic acid, compound with 2 - amino-2-(hydroxymethyl)-1,3-propanediol (1:1)
  • Inhibits prostaglandin synthesis
  • Biological activity associated with the S-form
  • Linear pharmacokinetic property
  • Highly protein bound
  • Metabolized in liver
  • Metabolites and some unchanged drugs excreted in urine

Pharmacology

  • Potent analgesic of the non-steroidal anti-inflammatory drugs (NSAIDs)
  • Inhibits the cyclooxygenase enzyme system
  • Demonstrates minimal anti-inflammatory effect at analgesic dose

Dosage & Administration

  • Recommended tablet dose: 10 mg every 4-6 hours, short-term use only (up to 7 days)
  • Injection may be used for moderate acute pain requiring analgesia at the opioid level

Interaction

  • Other NSAIDs or aspirin increase side effects
  • Anti-coagulants enhance anti-coagulant effect
  • Beta Blockers reduce anti-hypertensive effect
  • ACE Inhibitors increase risk of renal impairment
  • Methotrexate enhances the toxicity of methotrexate

Contraindications

  • Hypersensitivity to Ketorolac or other NSAIDs
  • Children under 16 years of age
  • Prophylactic analgesic before surgery
