Indications
- Management of moderate to severe acute post-operative pain
- Relief of pain, inflammation, and light sensitivity after ocular surgery
- Reduction of eye inflammation or redness caused by seasonal allergies
Description
- Belongs to pyrrolo-pyrrole group of nonsteroidal anti-inflammatory drugs (NSAID)
- Chemical name: 5 benzoyle-2,3-dihydro-1H-pyrroligine-1-carboxylic acid, compound with 2-amino-2-(hydroxymethyl)-1,3-propanediol (1:1)
- Inhibits synthesis of prostaglandins
- Peripherally acting analgesic
- Highly protein bound
- Metabolized in the liver
- Metabolites and some unchanged drug excreted in urine
Pharmacology
- Inhibits cyclooxygenase enzyme system
- Minimal anti-inflammatory effect at analgesic dose
Dosage & Administration
- Tablet: Recommended dose of 10 mg every 4-6 hours for short-term use (up to 7 days)
- Injection: Single or multiple doses for postoperative pain; IV bolus must be given over at least 15 seconds
- Eye Drops: 1 drop in each eye 4 times daily
- Specific dosing for different age groups and conditions
Interaction
- Increase in side effects with other NSAIDs or aspirin
- Enhanced anti-coagulant effect with anti-coagulants
- Reduction in anti-hypertensive effect with Beta Blocker
- Increased risk of renal impairment with ACE Inhibitors
- Enhanced toxicity of methotrexate
Contraindications
- Hypersensitivity to the drug or other NSAIDs
- Children under 16 years of age
- Prophylactic analgesic before surgery

Precautions & Warnings
- Caution in patients over 65 years old
- Caution in patients with active or suspected peptic ulcer, gastrointestinal bleeding, asthma, and liver dysfunction
- Eye Drops: Discontinue if evidence of corneal epithelial breakdown; should not be used while wearing contact lenses
