Description
- A medication to treat dyspeptic symptom complex
- Used for acute nausea and vomiting
- May be beneficial for Parkinson's disease

Dosage & Administration
- Should be taken 15-30 minutes before meals
- Usual recommended oral dose for adults is 10-20 mg every 6-8 hours daily
- Usual recommended oral dose for children is 2-4 ml suspension/10 kg body weight or 0.4-0.8 ml paediatric drops/10 kg body weight, every 6-8 hours daily
- Rectal administration for adults: 30-60 mg every 4-8 hours
Pharmacology
- Dopamine antagonist
- Blocks dopamine receptors in the Chemoreceptor Trigger Zone (CTZ) and stomach
- Restores normal motility and tone of the upper gastrointestinal tract
- Facilitates gastric emptying
- Enhances antral and duodenal peristalsis
Interaction
- Concomitant administration of anticholinergic drugs may antagonize the antidyspeptic effect of domperidone
- Antacids and antisecretory drugs should not be given simultaneously with domperidone as they lower its oral bioavailability
- In vitro data suggest that the concomitant use of drugs that significantly inhibit the CYP3A4 enzyme may result in increased plasma levels of domperidone
Contraindications
- Known hypersensitivity to the drug
- Neonates
- Gastrointestinal hemorrhage
- Mechanical obstruction or perforation
- Prolactin releasing pituitary tumor (prolactinoma)
Side Effects
- Transient intestinal cramps
- Extrapyramidal phenomena
- Increase in plasma prolactin level
- Rare allergic reactions such as rash and urticaria
Pregnancy & Lactation
- Should only be used during the first trimester of pregnancy if justified by the anticipated therapeutic benefit
- No evidence of increase in the risk of malformations in humans
- Not known whether excretion in breast milk is harmful to the newborn
Precautions & Warnings
- Use with caution in children due to increased risk of extrapyramidal reactions
- Use with caution in patients with hepatic impairment
Use in Special Populations
- Use in infants should be with great caution and under close medical supervision
- Use with caution in liver and kidney disorders
Overdose Effects
- Symptoms may include drowsiness, disorientation, and extrapyramidal reactions
- Recommendation for administration of activated charcoal and close patient observation
