Indications

  • Short-term management of moderate to severe acute post-operative pain
  • Relief of pain, inflammation, and light sensitivity after ocular surgery
  • Decreasing eye inflammation or redness caused by seasonal allergies

Description

  • Drug of pyrrolo-pyrrole group of nonsteroidal anti-inflammatory drug (NSAID)
  • Chemical composition: 5 benzoyle-2,3-dihydro-1H-pyrroligine-1-carboxylic acid, compound with 2 - amino-2-(hydroxymethyl)-1,3-propanediol (1:1)
  • Inhibition of prostaglandin synthesis
  • Pharmacokinetic property: linear, highly protein bound, largely metabolized in the liver
  • Excretion in the urine

Pharmacology

  • Potent analgesic of the non-steroidal anti-inflammatory drugs (NSAIDs)
  • Inhibition of cyclooxygenase enzyme system
  • Minimal anti-inflammatory effect at analgesic dose

Dosage & Administration

  • Tablet: Recommended dose is 10 mg every 4-6 hours, short-term use only
  • Injection: Administration guidelines for different age groups and conditions
  • Eye Drops: Administration guidelines for adults

Interaction

  • Increased side effects with other NSAIDs or aspirin
  • Enhanced anti-coagulant effect with anti-coagulants
  • Reduced anti-hypertensive effect with beta-blockers
  • Increased risk of renal impairment with ACE inhibitors
  • Enhanced toxicity of methotrexate

Contraindications

  • Hypersensitivity to Ketorolac or other NSAIDs
  • Use not recommended in children under 16 years of age
  • Contraindicated as prophylactic analgesic before surgery

  • Nausea, vomiting, gastro-intestinal bleeding, melena, peptic ulcer, pancreatitis
  • Anxiety, drowsiness, headache, excessive thirst, fatigue, bradycardia
  • Hypertension, palpitation, chest pain, infertility in females, pulmonary edema

Precautions & Warnings

  • Caution in patients over the age of 65 years
  • Caution in patients with active or suspected peptic ulcer or gastrointestinal bleeding, asthma, and liver dysfunction
  • Discontinuation of topical NSAIDs in the presence of corneal epithelial breakdown
  • Avoidance of administration while wearing contact lenses
