Indications

  • Short-term management of moderate to severe acute post-operative pain
  • Relief of pain, inflammation, and light sensitivity after ocular surgery
  • Decrease eye inflammation or redness caused by seasonal allergies

Description

  • Pyrrolo-pyrrole group of nonsteroidal anti-inflammatory drug (NSAID)
  • Chemical compound: 5 benzoyle-2,3-dihydro-1H-pyrroligine-1-carboxylic acid, compound with 2 - amino-2-(hydroxymethyl)-1,3-propanediol (1:1)
  • Inhibits synthesis of prostaglandins
  • Biological activity associated with the S-form
  • Highly protein bound
  • Metabolized in the liver
  • Metabolites and unchanged drugs excreted in urine

Pharmacology

  • Potent analgesic of the non-steroidal anti-inflammatory drugs (NSAIDs)
  • Inhibits the cyclooxygenase enzyme system
  • Minimal anti-inflammatory effect at analgesic dose

Dosage & Administration

  • Recommended dose for tablet: 10 mg every 4-6 hours
  • Not recommended for chronic use
  • Recommended dosage for injection in different age groups and conditions
  • Duration of analgesic effect for injection: 4 to 6 hours
  • Recommended dosage for eye drops: 4 times daily

Interaction

  • Increased side effects with other NSAIDs or aspirin
  • Enhanced anti-coagulant effect with anti-coagulants
  • Reduced anti-hypertensive effect with beta blockers
  • Increased risk of renal impairment with ACE inhibitors
  • Enhanced toxicity of methotrexate

Contraindications

  • Hypersensitivity to Ketorolac or other NSAIDs
  • Not for use in children under 16 years
  • Not to be used as prophylactic analgesic before surgery

  • Caution in patients with peptic ulcer, gastrointestinal bleeding, asthma, or liver dysfunction
  • Discontinue use of topical NSAIDs in patients with corneal epithelial breakdown
