Indications

  • Short-term management of moderate to severe acute post-operative pain

Description

  • Belongs to pyrrolo-pyrrole group of nonsteroidal anti-inflammatory drug (NSAID)
  • Chemical composition: 5 benzoyle-2,3-dihydro-1H-pyrroligine-1-carboxylic acid, compound with 2 - amino-2-(hydroxymethyl)-1,3-propanediol (1:1)
  • Inhibition of prostaglandin synthesis
  • Peripherally acting analgesic
  • Linear pharmacokinetic property
  • Highly protein bound
  • Metabolized in the liver
  • Excretion in the urine

Pharmacology

  • Potent analgesic
  • Inhibition of cyclooxygenase enzyme system

Dosage & Administration

  • Tablet: Recommended dose of 10 mg every 4-6 hours, recommended for short-term use (up to 7 days)
  • Injection: Use as single or multiple doses for management of moderately severe, acute pain
  • IV bolus given over no less than 15 seconds
  • IM administration given slowly and deeply into the muscle
  • Eye Drops: Recommended dose of 1 drop in each eye 4 times daily

Interaction

  • Increased side effects with other NSAIDs or aspirin
  • Enhanced anti-coagulant effect with anti-coagulants
  • Reduction in anti-hypertensive effect with Beta Blocker
  • Increased risk of renal impairment with ACE Inhibitors
  • Enhanced toxicity of methotrexate

Contraindications

  • Hypersensitivity to Ketorolac or other NSAIDs
  • Children under 16 years of age
  • Prophylactic analgesic use before surgery

Precautions & Warnings

  • Caution in patients with active or suspected peptic ulcer, gastrointestinal bleeding, asthma, and liver dysfunction
  • Discontinue use in case of corneal epithelial breakdown when using Eye Drops
