Indications

Description

  • Pyrrolo-pyrrole group of nonsteroidal anti-inflammatory drug (NSAID)
  • Chemical name: 5 benzoyle-2,3-dihydro-1H-pyrroligine-1-carboxylic acid, compound with 2-amino-2-(hydroxymethyl)-1,3-propanediol (1:1)
  • Inhibits synthesis of prostaglandins
  • Peripherally acting analgesic
  • Biological activity associated with the S-form
  • Linear pharmacokinetic property
  • Highly protein bound
  • Metabolized in the liver
  • Excreted in the urine

Pharmacology

  • Potent analgesic of the non-steroidal anti-inflammatory drugs (NSAIDs)
  • Inhibits cyclooxygenase enzyme system
  • Minimal anti-inflammatory effect at its analgesic dose

Dosage & Administration

  • Tablet: Recommended dose is 10 mg every 4-6 hours
  • Injection: Single or multiple doses for management of moderately severe, acute pain
  • Dosage varies based on age, weight, and route of administration
  • Eye Drops: Recommended administration frequency and dosage for adults

Interactions

  • Increased side effects with other NSAIDs or aspirin
  • Enhanced anti-coagulant effect with anti-coagulants
  • Reduced anti-hypertensive effect with beta blockers
  • Increased risk of renal impairment with ACE inhibitors
  • Enhanced toxicity of methotrexate

Contraindications

  • Hypersensitivity to drug or other NSAIDs
  • Children under 16 years of age
