Indications

Description
- Pyrrolo-pyrrole group of nonsteroidal anti-inflammatory drug (NSAID)
- Chemical name: 5 benzoyle-2,3-dihydro-1H-pyrroligine-1-carboxylic acid, compound with 2-amino-2-(hydroxymethyl)-1,3-propanediol (1:1)
- Inhibits synthesis of prostaglandins
- Peripherally acting analgesic
- Biological activity associated with the S-form
- Linear pharmacokinetic property
- Highly protein bound
- Metabolized in the liver
- Excreted in the urine
Pharmacology
- Potent analgesic of the non-steroidal anti-inflammatory drugs (NSAIDs)
- Inhibits cyclooxygenase enzyme system
- Minimal anti-inflammatory effect at its analgesic dose
Dosage & Administration
- Tablet: Recommended dose is 10 mg every 4-6 hours
- Injection: Single or multiple doses for management of moderately severe, acute pain
- Dosage varies based on age, weight, and route of administration
- Eye Drops: Recommended administration frequency and dosage for adults
Interactions
- Increased side effects with other NSAIDs or aspirin
- Enhanced anti-coagulant effect with anti-coagulants
- Reduced anti-hypertensive effect with beta blockers
- Increased risk of renal impairment with ACE inhibitors
- Enhanced toxicity of methotrexate
Contraindications
- Hypersensitivity to drug or other NSAIDs
