Indications
- Short-term management of moderate to severe acute post-operative pain
- Relieving pain, inflammation, and light sensitivity after ocular surgery
- Decreasing eye inflammation or redness caused by seasonal allergies
Description
- Belongs to the pyrrolo-pyrrole group of nonsteroidal anti-inflammatory drugs (NSAID)
- Chemical structure: 5 benzoyle-2,3-dihydro-1H-pyrroligine-1-carboxylic acid, compound with 2-amino-2-(hydroxymethyl)-1,3-propanediol (1:1)
- Inhibits synthesis of prostaglandins
- The biological activity is associated with the S-form
- Pharmacokinetic properties: linear, highly protein-bound, largely metabolized in liver, excretion in urine
Pharmacology
- Potent analgesic of the non-steroidal anti-inflammatory drugs (NSAIDs)
- Acts by inhibiting the cyclooxygenase enzyme system and inhibits prostaglandin synthesis
- Minimal anti-inflammatory effect at analgesic dose
Dosage & Administration
- Tablet recommended dose: 10 mg every 4-6 hours for short-term use (up to 7 days)
- Injection for moderate to severe acute pain management, with specific dosing based on age and condition
- Eye Drops: 1 drop in each eye 4 times daily
Interaction
- Increased side effects with other NSAIDs or aspirin
- Enhanced anti-coagulant effect with anti-coagulants
- Reduced anti-hypertensive effect with beta blockers
- Increased risk of renal impairment with ACE inhibitors
- Enhanced toxicity of methotrexate
Contraindications
- Hypersensitivity to ketorolac or other NSAIDs
- Children under 16 years of age
- Prophylactic analgesic use before surgery

Precautions & Warnings
- Caution in patients over 65 years of age
- Caution in patients with active or suspected peptic ulcer or gastrointestinal bleeding, asthma, and liver dysfunction
- Discontinue use of topical NSAIDs and monitor corneal health in patients with evidence of corneal epithelial breakdown
- Avoid administration while wearing contact lenses for ophthalmic solution
