Description
- Belongs to pyrrolo-pyrrole group of nonsteroidal anti-inflammatory drugs (NSAID)
- Chemical composition: 5 benzoyle-2,3-dihydro-1H-pyrroligine-1-carboxylic acid, compound with 2 - amino-2-(hydroxymethyl)-1,3-propanediol (1:1)
- Inhibits synthesis of prostaglandins and acts as a peripherally acting analgesic
- Pharmacokinetic property is linear
- Highly protein-bound and largely metabolized in the liver
- Metabolites and some unchanged drugs are excreted in urine
Mode of Action
- Potent analgesic of the non-steroidal anti-inflammatory drugs (NSAIDs)
- Inhibits the cyclooxygenase enzyme system and prostaglandin synthesis
- Minimal anti-inflammatory effect at analgesic dose
Dosage & Administration
- Tablet: Recommended dose is 10 mg every 4-6 hours for short-term use (up to 7 days)
- Tablet: Doses exceeding 40 mg/day is not recommended
- Injection: Used for the management of moderately severe acute pain, with specific dosage guidelines for different age groups
- Eye Drops: Recommended dosage is 1 drop in each eye 4 times daily
Interaction
- Increases side effects with other NSAIDs or aspirin
- Enhances anti-coagulant effect of anti-coagulants
- Reduces the anti-hypertensive effect of Beta Blockers
- Increases the risk of renal impairment with ACE Inhibitors
- Enhances the toxicity of methotrexate
Contraindications
- Hypersensitivity to Ketorolac or other NSAIDs
- Use is contraindicated in children under 16 years of age
- Contraindicated as prophylactic analgesic before surgery

- Take caution in patients with active or suspected peptic ulcer, gastrointestinal bleeding, asthma, or liver dysfunction
- Discontinue use if evidence of corneal epithelial breakdown is found in patients using eye drops
