Description

  • Belongs to pyrrolo-pyrrole group of nonsteroidal anti-inflammatory drugs (NSAID)
  • Chemical composition: 5 benzoyle-2,3-dihydro-1H-pyrroligine-1-carboxylic acid, compound with 2 - amino-2-(hydroxymethyl)-1,3-propanediol (1:1)
  • Inhibits synthesis of prostaglandins and acts as a peripherally acting analgesic
  • Pharmacokinetic property is linear
  • Highly protein-bound and largely metabolized in the liver
  • Metabolites and some unchanged drugs are excreted in urine

Mode of Action

  • Potent analgesic of the non-steroidal anti-inflammatory drugs (NSAIDs)
  • Inhibits the cyclooxygenase enzyme system and prostaglandin synthesis
  • Minimal anti-inflammatory effect at analgesic dose

Dosage & Administration

  • Tablet: Recommended dose is 10 mg every 4-6 hours for short-term use (up to 7 days)
  • Tablet: Doses exceeding 40 mg/day is not recommended
  • Injection: Used for the management of moderately severe acute pain, with specific dosage guidelines for different age groups
  • Eye Drops: Recommended dosage is 1 drop in each eye 4 times daily

Interaction

  • Increases side effects with other NSAIDs or aspirin
  • Enhances anti-coagulant effect of anti-coagulants
  • Reduces the anti-hypertensive effect of Beta Blockers
  • Increases the risk of renal impairment with ACE Inhibitors
  • Enhances the toxicity of methotrexate

Contraindications

  • Hypersensitivity to Ketorolac or other NSAIDs
  • Use is contraindicated in children under 16 years of age
  • Contraindicated as prophylactic analgesic before surgery

  • Discontinue use if evidence of corneal epithelial breakdown is found in patients using eye drops
